Resveratrol induces autophagy in endometrial cancer Ishikawa cells by inhibiting the PI3K/AKT pathway

〔Abstract〕 Objective To explore the mechanism of resveratrol in promoting autophagy of endometrial cancer Ishikawa cells. Methods Ishikawa cells were divided into three groups: control, resveratrol and resveratrol+PI3 Kagonist(740 Y-P).The expression levels of LC3,Beclin1,PI3 K and AKT were detected by qRT-PCR and Western Blot method, the expression intensity of LC3 was detected by cell immunofluorescence method, and the autophagy of cells was observed by electron microscope. Results Resveratrol could increase the mRNA and protein levels of Beclin1 and LC3 in cells; the red fluorescence intensity of cytoplasmic LC3 was enhanced; the number of autophagosomes increased.The P-PI3 K and P-AKT protein levels of Ishikawa cells in the resveratrol group were lower than those in the control group, while the autophagy level of the resveratrol+740 Y-P group was lower than that in the resveratrol group. Conclusion Resveratrol may induce autophagy in endometrial cancer Ishikawa cells by inhibiting PI3 K/AKT pathway, thereby inhibiting cell proliferation.Resveratrol may become a new drug for adjuvant treatment of endometrial cancer.
