Long non coding RNA-SNHG4 promotes cell proliferation and mediates cisplatin resistance in gastric cancer

〔Abstract〕 Objective To establish lncRNA-SNHG4 overexpressing gastric cancer cell lines; to examine the role of SNHG4 on cell growth, proliferation and the sensitivity to cisplatin in gastric cancer cells; to compare the expression levels of SNHG4 in gastric cancer and adjacent normal tissues, and to analyze the correlation between SNHG4 expression levels and pathological characteristics in gastric cancer patients. Methods The expression levels of SNHG4 in cell lines and tissues were detected by RT-qPCR; cell viabilities, cell proliferation and growth ability were detected by MTT and 3-D cell culture assay respectively; the correlation between SNHG4 expression levels and pathological characteristics in gastric cancer patients was analyzed by chi square test. Results MTT and 3-D cell culture experiments showed that overexpression of SNHG4 promoted the proliferation and cisplatin resistance in gastric cancer cell lines MKN-45 and AGS(P<0.05); the expression levels of SNHG4 in gastric cancer tissues were significantly higher than that in adjacent normal tissues; the high expression levels of SNHG4 in gastric cancer tissues were significantly correlated with tumor volume, lymph node metastasis and pathological grade in gastric cancer patients(P<0.05), but not with patients′age, gender or clinical stage. Conclusion SNHG4 promotes cell proliferation, growth and cisplatin resistance in gastric cancer cells. The high expression levels of SNHG4 are correlated with pathological characteristics in gastric cancer patients. SNHG4 has important clinical significance in gastric cancer.
