〔Abstract〕 Objective To investigate the interaction of JAK2/STAT3 pathway and transient receptor potential channel 6(TRPC6) signaling path way in Podocyte induced by Angiotensin Ⅱ(Ang Ⅱ). Methods Podocyte in mice was treated by the Ang Ⅱ, Ang Ⅱ+chlorine mitotane, Ang Ⅱ+AG490, and Ang Ⅱ+SKF96365. The expression levels of TRPC6, JAK2/STAT3 protein and mRNA were detected by Western blot and qRT-PCR. The secretion of tumor necrosis factor α(TNF-α) and interleukin 6(IL-6) was detected by ELISA. Results The expression of JAK2/STAT3, TRPC6、TNF-α and IL-6 in Ang Ⅱ injury group was higher than those in control group(P<0.05). The expression of JAK2/STAT3, TRPC6、TNF-α and IL-6 in Ang Ⅱ+chlorine mitotane group was lower than those in Ang Ⅱ injury group(P<0.05). The expression of JAK2/STAT3, TRPC6、TNF-α and IL-6 in Ang Ⅱ+AG490 group was lower than those in Ang Ⅱ injury group(P<0.05). The expression of JAK2/STAT3, TRPC6、TNF-α and IL-6 in Ang Ⅱ+SKF96365 group was lower than those in Ang Ⅱ injury group(P<0.05). Conclusion Ang Ⅱ can activate JAK2/STAT3 pathway by TRPC6, and the expression of TRPC6 can be cut by the inhibition of JAK2/STAT3. There is a positive feedback mechanism between JAK2/STAT3 and TRPC6.
