Effect of CLIC4 on the proliferation and migration of gastric cancer SGC-7901 cells

〔Abstract〕 Objective To investigate the effects of chloride intracellular channel 4(CLIC4) on the proliferation and migration ability of gastric cancer cell line SGC-7901. Methods Lentiviral plasmids with CLIC4 shRNA were transfected into HEK 293 T cells and the supernatant with virus particles were collected and infected into SGC-7901 cells to construct stable CLIC4 low-expression cell lines with puromycin selection, and the protein levels of CLIC4 was detected by Western blot. Colony formation assays and CCK-8 assays were conducted for the proliferation of SGC-7901 cells after CLIC4 knockdown. The expressions of epithelial-mesenchymal transition( EMT)-related proteins such as E-cadherin and Vimentin were checked by Western blot in SGC-7901 cells after CLIC4 knockdown.Results A stable CLIC4 low-expressing SGC-7901 cell line was successfully constructed. The expression of CLIC4 did not affect the proliferation of SGC-7901 cells,while the EMT associated protein E-cadherin was up-regulated and Vimentin was down-regulated with CLIC4 knockdown. Conclusion CLIC4 does not affect the proliferation but promote the migration capacity of SGC-7901 cells.
