〔Abstract〕 Objective To investigate the effect of MAST3 on proliferation and migration in SGC-7901 cells.Methods Western blot and qRT-PCR were used to detect MAST3 expression in gastric cancer tissues. MAST3-RNAi and p Ex-3-MAST3 were further transfected into SGC-7901 cells,and the expressions of CyclinD1,C-myc,MMP9 and MMP3 were detected by qRT-PCR and Western blot,respectively,and the effect of MAST3 on the proliferation and migration of SGC-7901 cells was observed. Cell cycle was detected by flow cytometry. Results Western blot results showed that MAST3 was significantly higher in gastric cancer tissues than that in adjacent tissues( P<0. 01),and the expression increased significantly after TGF-β1 stimulation. After MAST3-RNAi inhibited the expression of MAST3,the expression of CyclinD1 and C-myc protein was significantly lower than that of the control group( P<0. 01),and the cells in S phase and G2/M phase significantly decreased,while the expression of MMP-9 and MMP-3 protein was also significantly lower than that of the control. The expression of CyclinD1 and Cmyc protein was significantly higher than that of the control group after p Ex-3-MAST3 over-expression of MAST3( P<0. 01),and the cells in S phase and G2/M phase significantly increased,MMP-9 and MMP-3 protein expression was also significantly higher than of the control group. Conclusion MAST3 can regulate the proliferation and migration of gastric cancer SGC-7901 cells.
