miR-223-3p promotes osteogenic differentiation of dental pulp stem cells by inhibiting VHL

〔Abstract〕 Objective To investigate the effect of miR-223-3 p on osteogenic differentiation of human dental pulp stem cells(hDPSCs) by inhibiting Von Hippel-Lindau(VHL) gene. Methods Hypoxia induced hDPSCs for 24 h(oxygen concentration 1%).The changes of cell proliferation were observed by MTT assay.The miRNA of miR-223-3 p was predicted by bioinformatics, and the relationship between miR-223-3 p and VHL was verified by luciferase.The expression of miR-223-3 p mimics or miR-223-3 p was transfected into DPSCs, and the expression of miR-223-3 p, VHL, hypoxia inducible factor-1α(HIF-1α),runt-related transcription factor 2(Runx2), bone morphogenetic protein 2(BMP2) mRNA, VHL, HIF-1α was detected by qRT-PCR and Western blot. Results miR-223-3 p increased in the hypoxic-induced DPSCs(P<0.05).The MTT results showed that miR-223-3 p mimics promoted the proliferation of DPSCs(P<0.05).Moreover, miR-223-3 p regulated the expression level of VHL by targeting VHL 3′-UTR.The transfection of miR-223-3 p was detected by qRT-PCR and Western blot, which significantly promoted the expression of HIF-1α, Runx2 and BMP2 genes(P<0.05). Conclusion miR-223-3 p promotes osteogenic differentiation of dental pulp stem cells by inhibiting VHL gene.
