〔Abstract〕 Objective To elucidate the role of long non-coding RNA(lncRNA), a novel tumor regulator, PVT1 in hepatocellular carcinoma(HCC). Methods According to the analysis of TCGA database, PVT1 was highly expressed in HCC. Transient transfection was used to construct PVT1 knockdown cell lines. CCK-8 assay and scratch assay were used to evaluate the ability of cell growth and metastasis. Bioinformatics prediction and luciferase assay were used to validate the targeted binding of PVT1 to miR-17-5 p. The co-transfection experiment was used to verify the effect of PVT1 on proliferation and metastasis through miR-17-5 p. Results The results showed that PVT1 was highly expressed in HCC tissues and cell lines. Knockdown of PVT1 decreased the ability of cells proliferation and metastasis. Luciferase assay confirmed that PVT1 could bind to miR-17-5 p, and miR-17-5 p mediated the effect of PVT1 on the proliferation and metastasis of HCC cells. Conclusion lncRNA PVT1 can enhance the proliferation and metastasis of HCC cells through miR-17-5 p. The above results indicate the important role of PVT1 in HCC and provide a good prospect for future targeted therapy.
