〔Abstract〕 Objective By doping Cu2+with mesoporous silica, a copper silicate nano platform with chemical kinetic effect was prepared, which can catalyze hydrogen peroxide to produce strong oxidative hydroxyl radicals, realize the oxidative damage of cancer cells and lead to cancer cells death.Methods By hydrothermal method, mesoporous silica was etched by copper ion to prepare copper silicate nanoparticles.The morphology, crystal structure, porosity, specific surface area and other physical and chemical properties of the nanoparticles were characterized by transmission electron microscope, X-ray diffraction and nitrogen adsorption desorption curve.Secondly, the biocompatibility of mesoporous silica without copper ions was verified by cell experiments.At the same time, the number of cytotoxic hydroxyl radicals produced by the copper silicate nanoparticles in cancer cells was evaluated by cell experiments,and the therapeutic effect of the nanoparticles on cancer cells was further evaluated.Results TEM images showed that mesoporous silica and copper silicate nanoparticles showed spherical morphology with good monodispersity.The average hydration particle sizes were 85.9 nm and 204.4 nm,respectively.The results of nitrogen adsorption desorption curve showed that the specific surface area of mesoporous silica and copper silicate nanoparticles were 26.8 m2/g and 5.36 m2/g,respectively,and the pore size of the nanoparticles was 18.9 nm and 1.66 nm,respectively.Subsequently,through cell experiments,we knowed that mesoporous silica nanoparticles had excellent biocompatibility,while copper silicate nanoparticles showed a strong inhibition on cancer cells.Conclusion In this study,copper silicate nanoparticles were successfully prepared and achieved good antitumor performance,indicating that they have a good prospect of chemokinetic therapy.
