Differential proteomics and bioinformatics analysis of small intestine of atherosclerosis Bama pig with spleen deficiency and phlegm turbid under TCM intervention

〔Abstract〕 Objective To study the effect of TCM intervention on differential proteomics of small intestine in atherosclerosis(AS) bama pig with spleen deficiency and phlegm turbid by iTRAQ technique. Methods Ten healthy half-year-old male Guangxi Bama miniature pigs were randomly divided into model group and TCM group, five pigs in each group. Total proteins of small intestine was isolated, labeled with iTRAQ reagent and detected by mass spectrometry. The differentially expressed proteins were analyzed by software, and the proteomics differences between the model group and the treatment group were analyzed by LC-ESI-MS/MS method. The significantly enriched proteins were selected GO function annotation, enrichment and KEGG pathway enrichment. Results A total of 189 different proteins were screened in the treatment group compared with the model group, including 70 up-regulated proteins and 119 down-regulated proteins. GO enrichment analysis showed that these differential proteins were mainly concentrated in biological processes such as binding, catalytic activity, transport activity and structural molecular activity. It was concentrated in cells, cell components, organelles, cell membranes and extracellular regions. The molecular functions such as cell process, biological regulation, biological process regulation, metabolic process, stimulation response and localization were enriched.Analysis of the KEGG pathway showed that cell adhesion molecular pathways had the most significant changes, with significant changes in phagocytosis, prominent vesicle circulation, amino acid biosynthesis, insulin secretion, PPAR signaling pathways, fat digestion and absorption, and bile acid secretion pathways closely related to lipid metabolism.In addition, oxidative phosphorylation and other pathway proteins closely related to energy metabolism also changed. Conclusion The differential proteomics changes of small intestine in bama pig with spleen deficiency and phlegm turbid under the intervention of TCM may be closely related to cellular inflammatory response, lipid metabolism and bile acid secretion.Intestinal digestion and absorption in the state of spleen deficiency and high fat are abnormal, and the cellular inflammatory reaction may be closely related to the formation of AS. The therapeutic targets of TCM intervention may be in the above directions.
