abstract:
Objective To establish a stable and reliable mouse model ofKlebsiella pneumoniaeliver abscess(KPLA) for deep research. Methods The clinical isolates ofKlebsiella pneumoniae(KP)181608 were selected as the experimental strains. Mice were intraperitoneally injected with bacterial suspensions of different concentrations. Lethal dose 50(LD50) was calculated by the modified coon method, and the concentration of liver abscess model mice was 1/2 LD50. The general condition, weight change, blood routine examination, HE staining and liver bacterial burden were recorded to comprehensively evaluate the model and verify its effect. Results The LD50of KP was 104CFU/ml according to the improved Karber′s method. The result of bacterial culture from the site of liver abscess was positive, and the infected bacteria was identified as KP by colony morphology, positive wire-drawing test and mass spectrometry. The results of HE staining suggested large area of necrosis and infiltration of inflammatory cells in liver of KLA mice. With the extension of the infection time, the mental state of KLA mice became worse, coupling with the decrease of food intake, eventually the weight obviously decreased. The number of leukocytes in infected mice were higher than that in the control group, and the difference was statistically significant(P<0.05). In addition, alanine aminotransferase(ALT),aspartate aminotransferase(AST) and inflammatory factors [interleukin(IL)-1β,IL-6 and tumor necrosis factor-α(TNF-α)] significantly increased in KLA group, and RT-qPCR results also showed higher mRNA levels of inflammatory and chemokine factors [IL-1β,IL-6,IL-10 and monocyte chemotactic protein-1(MCP-1)]than those of the control group, and the difference was statistically significant(P<0.05). Conclusion The KPLA mouse is successfully established by intraperitoneal injection, providing a stable and easy-to-replicate animal model of liver abscess for further study.

Objective To explore the function of miR-27 a-3 p on osteoblast differentiation of hDPSCs by targeting Yes-related protein 1(YAP1). Methods Human dental pulp stem cells(hDPSCs) were culturedin vitroand transfected with miR-27 a-3 p mimics(miR-27 a-3 p overexpression group), miR-27 a-3 p inhibitor(miR-27 a-3 p knock-down group) and miR-NC(control group), respectively. Real-time PCR(qRT-PCR) and Western blot were used to detect the effect of miR-27 a-3 p expression levels on YAP1. Bioinformatics predicted the target gene of miR-27 a-3 p which was then verified by dual luciferase reporter gene experiments. In addition, miR-27 a-3 p mimics and miR-NC were transfected into hDPSCs at the time stated. The expression of miR-27 a-3 p and the biochemical markers of bone formation of hDPSCs, such as bone morphogenetic protein-2(BMP-2) and runt-related transcription factor 2(RUNX2) and osteopontin(OPN), YAP1 mRNA was detected by qRT-PCR. Western blot was applied to detect the expression of BMP-2, RUNX2, OPN and YAP1 protein. The effect of miR-27 a-3 p or YAP1 on the proliferation of hDPSCs was analyzed by MTT assay. Results The expression of miR-27 a-3 p in the miR-27 a-3 p overexpression group increased in comparison with the control group(P<0.05), when that of YAP1 mRNA and protein was suppressed(P<0.05). Compared with control group, the expression of miR-27 a-3 p in miR-27 a-3 p knock-down group decreased(P<0.05) as that of YAP1 mRNA and protein increased(P<0.05). The target gene of miR-27 a-3 p was YAP1, which was predicted by bioinformatics and verified by dual luciferase reporter gene experiments(P<0.05). Overexpression of miR-27 a-3 p could promote expression of BMP-2, RUNX2, OPN mRNA and protein(P<0.05) while down-regulating the expression of YAP1 mRNA and protein. Overexpression of YAP1 could mitigate the effect of miR-27 a-3 p on proliferation of hDPSCs(P<0.05). Conclusion miR-27 a-3 p promotes osteogenic differentiation of hDPSCs, which may be related to the down-regulation of YAP1 by miR-27 a-3 p.

Objective To explore the function of Slit2/Robo4/Rho signaling pathway in monocrotaline(MCT)-induced hepatic sinusoidal obstruction syndrome(HSOS). Methods Thirty-four rats were randomly divided into control group and experimental group. The rats in experimental group was established with MCT by gastric lavage(160 mg/kg), and randomly sacrificed after model making for 1 day, 2 days and 4 days. Serum alanine aminotransferase(ALT) and aspartate aminotransferase(AST) were measured by chemical method.Liver tissue damage was observed by HE staining and Masson staining. α-smooth muscle actin(α-SMA) protein expression in rat liver was measured by immunohistochemistry. The related indexes of Slit2/Robo4/Rho signaling pathway were measured by Western blot and RT-qPCR. Results After a single intragastric administration of MCT(160 mg/kg),the congestion of hepatic sinusoid, the degeneration of hepatocyte and central venous endothelial cell injury of the 1-day group were observed, the congestion and dilatation of hepatic sinusoid of the 2-day group was obvious, hepatic pathological changes were more serious of the 4-day group and perisinusoidal fibrosis was seen in the 4-day group. Compared with the control group, the liver index of the experimental group increased(P<0.05) serum AST of the 2-day group and the 4-day group increased(P<0.01), serum ALT of the 2-day group increased(P<0.01). α-SMA protein expression in rat liver of all treatment group was significantly higher than the control group(P<0.01). The expressions of Slit2, Robo4 mRNA and protein in each experimental group was lower than those of the control group(P<0.01) and gradually decreased with the prolonged administration time(P<0.05), while the expressions of RhoA,Rac1,Cdc42,matrix metalloproteinase(MMP)-2, MMP-9 mRNA and protein in each experimental group were higher than those of the control group(P<0.01) and gradually increased(P<0.05). Conclusion Rat HSOS was induced by single MCT, and rat liver injury was progressively aggravated. Slit2/Robo4/Rho signaling pathway, hepatic stellate cell activation and increase of MMP-2 and MMP-9 may be one of the important mechanisms of HSOS induced by MCT.

Effect of lentivirus-mediated PDIA3 overexpression on intestinal mucosal barrier in mice with ulcerative colitis and its mechanism

abstract:
Objective To investigate the protective effect of protein disulfide isomerase A3(PDIA3) on ulcerative colitis(UC) mice and its possible mechanism. Methods Sixty C57 BL/6 mice were randomly divided into a normal Control group(Control), Model group(Model), no-load lentivirus group(Lv-NC) and a PDIA3 overexpressed lentivirus group(Lv-PDIA3), with 15 mice in each group. The mouse model of UC was established by drinking 3% dextran sulfate sodium(DSS) solution for 7 consecutive days. After successful modeling, lentivirus intervention was started. After 7 days of lentivirus intervention, the mice in each group were evaluated by disease activity index(DAI) score. The histopathological changes of colon were observed by HE staining. The inflammatory cytokines of interleukin 1β(IL-1β),interleukin 6(IL-6),tumor necrosis factor α(TNF-α), and the intestinal mucosal barrier permeability markers of D-lactic acid(D-Lac), diamine oxidase(DAO) levels in serum were detected by ELISA, PDIA3 mRNA levels in colon tissues were detected by qRT-PCR. The expression levels of PDIA3, ZO-1, Occludin, Claudin-1, IκBα, p-NF-κB p65(Ser536) and NF-κB p65 in colonic tissue were detected by Western blot. Results Compared with the Control group, the colon tissue of the Model group was severely damaged. DAI score, the levels of IL-1β, IL-6, TNF-α, D-Lac, DAO in serum, and the expression levels of IκBα and p-NF-κB p65(Ser536) in colon tissues were increased(P<0.01). The proteins expression levels of ZO-1, Occludin and Claudin-1 in colon tissues were reduced(P<0.01). Compared with the Model group, the damage degree of colonic tissue in Lv-PDIA3 group was reduced. DAI score, the levels of IL-1β, IL-6, TNF-α, D-Lac, DAO in serum, and the expression levels of IκBα and p-NF-κB p65(Ser536) in colon tissues were decreased(P<0.01). The proteins expression levels of ZO-1, Occludin and Claudin-1 in colon tissues were increased(P<0.01). However, there was no significant difference in Lv-NC group(P>0.05). Conclusion Overexpression of PDIA3 improved the impaired intestinal mucosal barrier function in UC mice, which may be related to inhibition of NF-κB signaling pathway activation.

abstract:
Objective To explore the regulation of the co-signaling molecule B7-H3 on the expression of interleukin 22(IL-22) and its clinical significance in lung adenocarcinoma. Methods CRISPR/Cas9 technology was used to knock out the B7-H3 gene of lung adenocarcinoma A549 cells(A549/KO), meanwhile, serum of 56 patients with lung adenocarcinoma was collected. Enzyme linked immunosorbent assay(ELISA) was used to detect the A549/KO culture supernatation and IL-22 level in serum of the patients and the content of soluble B7-H3(sB7-H3) in serum of the patients. The correlation between sB7-H3 and IL-22 was analyzed, the Logistic regression model was established to predict the risk factors of lung adenocarcinoma, and the value of sB7-H3 and IL-22 in the diagnosis of lung adenocarcinoma was discussed using receiver operating characteristic curve(ROC). Results The expression level of IL-22 in the supernatant of A549/KO cells was significantly lower than that of wild-type A549(P<0.05). The expression levels of sB7-H3 and IL-22 in the serum of patients with lung adenocarcinoma were significantly higher than those of healthy controls(P<0.05). The expression levels of sB7-H3 and IL-22 in the serum of patients with lung adenocarcinoma were related to pT stage, lymph node metastasis and pTNM stage(allP<0.05), and the expression levels of sB7-H3 and IL-22 were positively correlated(r s =0.316,P<0.05). sB7-H3(OR=1.423,95%CI: 1.139-1.778) and IL-22(OR=1.108,95%CI: 1.001-1.228) were selected as a predictors of lung adenocarcinoma through multivariate Logistic regression analysis. As a diagnostic indicator of lung adenocarcinoma. ROC curve analysis showed that the combined detection of IL-22 and sB7-H3(AUC=0.827,95%CI: 0.727-0.903) was better than the detection of sB7-H3 alone(AUC=0.795, 95%CI: 0.690-0.877) and IL-22 was tested alone(AUC=0.766, 95%CI: 0.658-0.853). Conclusion B7-H3 can play a role in the occurrence and development of lung adenocarcinoma by up-regulating IL-22 expression. Combined detection of serum sB7-H3 and IL-22 is helpful for the diagnosis and monitoring of lung adenocarcinoma.

abstract:
Objective To analyze the clinical and biochemical indicators and their distribution characteristics of recurrent spontaneous abortion(RSA) patients to explore the relationship between RSA and obesity, insulin resistance and abnormal glucose metabolism. Methods 876 women who suffered from RSA were recruited as the case group(RSA group), and 1 397 women who underwent assisted reproductive technology due to male factors in the same period were selected as the control group. The clinical data were collected, and the body mass index(BMI),fasting insulin( FINS),fasting plasma glucose( FPG) levels of the two groups were analyzed. At the same time,the BMI level was divided into 4 intervals,and the distribution characteristics of insulin and blood glucose in the two groups under different intervals were compared. Then the variables with statistically significant differences were selected for Logistic regression analysis. Results The FPG,FINS and HOMA-IR indexes of the RSA group were higher than those of the control group( P<0. 01); The detection rates of impaired fasting blood glucose( IFG) and insulin resistance( IR) in the RSA group were higher than those in the control group( P<0. 05). The occurrence rate of abnormal BMI and glucose metabolism in the RSA group was higher than that in the control group( P<0. 05). The generation rate of abnormal glucose metabolism in overweight and obese patients in the RSA group was higher than that in the control group( P<0. 05); the detection rate of IR in normal weight patients in the RSA group was higher than that in the control group( P<0. 01). Multivariate analysis showed that IR was a risk factor for RSA. Conclusion The occurrence of RSA is related to IR and abnormal glucose metabolism. Overweight or obese patients with RSA are more likely to have glucose metabolism disorders,and attention should be paid to the endocrine metabolism status of women with RSA of normal weight.
